Reporter gene expression driven by the HCV IRES is sensitive to inhibition during apoptosis. Huh-7 hepatoma cells stably expressing a Renilla luciferase-HCV IRES-firefly luciferase bicistronic construct were incubated with or without TRAIL (0.5 μg/mL) for 6 h or 22 h to induce apoptosis. Cell extracts were prepared and the luciferase activities were determined as described in Materials and Methods. (A) Cap-dependent Renilla luciferase and IRES-dependent firefly luciferase activities.
