Mutations that alter levels of Arg8p reporter protein do not alter steady-state mRNA levels. The top two panels show the results of sequential probing of a Northern blot containing 10 μg of total RNA per sample. The blot was probed first with a radioactively labeled probe against ARG8m (top) and then with a probe against the mitochondrial 15S rRNA (middle) as a loading control. The third panel shows the results of probing a Western blot containing 75 μg of total protein per sample with an antibody against Arg8p. The COX2 negative control or cox2::ARG8m alleles (shown according to the COX2 codons present in the fusion construct) are as written above the top panel, and the respective growth phenotypes are as shown at the bottom of the figure. Strains shown, from left to right: EHW154, NB80, EHW155, and EHW195–EHW204 (Table 1).
