Protein ligands mediate the CRM1-dependent export of HuR in response to heat shock

Efforts to reproduce certain of the results in Figure 6 of the paper cited above have not been successful. Microscopic examination had given the impression of nuclear accumulation of mRNA (panels A1–4), but this apparent effect was probably a consequence of the fact that many cells round up after heat shock. In subsequent biochemical experiments, we have so far observed no inhibition of GAPDH mRNA export to the cytoplasm after heat shock, and slight rather than complete nuclear retention of hsp70 mRNA induced by leptomycin B (panel B).

Therefore the conclusions originally derived from these panels—(1) that most mRNAs are retained in the nucleus of HeLa cells after heat shock because of an export block, and (2) that hsp70 mRNA exits the nucleus after heat-induced stress primarily via a CRM1-dependent pathway—are not supported by our additional experiments.
