The assembly and activity of the eIF4F complex are regulated by the mTOR and MNK1/2 kinases, which phosphorylate 4E-BPs and eIF4E, respectively. Many inhibitors have been developed that target RAS/MEK/ERK and PI3K/Akt/mTOR pathways in cancer (Montagut and Settleman 2009; Hua et al. 2019; He et al. 2021).
