TLC analyses reveal adenosine-to-inosine (A-to-I) editing. Two-dimensional TLC analyses using four different dsRNAs, each containing one strand that was labeled during in vitro transcription with 32P-ATP (A), 32P-CTP (C), 32P-GTP (G), or 32P-UTP (U). dsRNAs were reacted for 1 h in a X. laevis embryo nuclear extract (+), or a heat-inactivated embryo nuclear extract (−). After incubation, reactions were deproteinized, digested to nucleotides with nuclease Pl, and each sample, as well as a mixture of both (+/−), were spotted at the bottom left-hand corner of a cellulose chromatography plate. Directions of chromatography are indicated by small arrows at the plate origins, with the first dimension indicated by the vertical arrow and the second dimension indicated by the horizontal arrow.
