RNA capping activity of FCE and VCE on FLuc transcript (1.8 kb) containing a 5′sequence derived from the first 50 nt of human β globin variant 1, synthetic 5′ UTR of pRNA21, synthetic 5′ UTR of pFLuc, and Comirnaty. FCE or VCE (20 nM) was used to cap 3.5 µM of the in vitro transcripts for 1 h at 37°C. Following capping reactions, mRNA capping was measured using targeted RNase H cleavage and LC–MS.
