(A) Guide RNAs with the ATATA initiation sequence followed by an 11–12 nt anchor fit just inside the molecular ruler (blue shape), which allows editing of editing sites (“u”s in the magenta ellipse). (B) The 5′ end of anchors of gRNAs whose anchors completely overlap their initiation sequences (almost always AAAAB or AAABN) would be buried within the ruler, thereby preventing editing of editing sites (e.g., between C, A, and G on the mRNA). (C) This is compensated for by the ∼5 nt oligo(A) tails that are attached to such gRNAs (Fig. 8B).
