Wobble uridine tRNA modifications and ALKBH8 activities. (A) Known wobble uridine modifications in yeast. (B) The ALKBH8 reaction mechanism. The MTase domain of ALKBH8 together with the cofactor TRMT112 uses S-adenosyl-L-methionine (SAM) to convert cm5U into mcm5U. In the case of tRNAGly(UCC), the AlkB-like domain subsequently performs hydroxylation into (S)-mchm5U.
