(A) Trans-translation of sfGFP1-10 mRNA lacking a stop codon. The tmRNAGFP11-SmpB complex binds to the stalled ribosome, and translation resumes thanks to the tmRNAGFP11 mRNA-like domain (MLD). The MLD encodes the missing eleventh beta-strand of the sfGFP, and the complete sfGFP is released and becomes fluorescent. (B) Impairment of the process in the presence of trans-translation inhibitors. The ribosomes stay stalled on the problematic mRNA and fluorescence is impaired.
