Species-specific tRNA processing intermediates under stress conditions: tRNAUAUIle contains the 5′ leader sequence whereas the aberrant tRNATyr species is processed at the 5′ end. Wild-type and rex1Δ mutant strains were grown in YPD at 30°C and then shifted for 2 h to 37°C and/or YPGly. RNA was isolated and analyzed by hybridization with probes specific to 5′ leaders of both tRNAUAUIle genes (A). and tRNATyr tY(GUA)D (B). Forms of tRNA precursors that accumulate under stress conditions are indicated by solid arrows. Hybridization with other specific probes recognizing mature and pre-tRNAs were employed as controls.
