
Mutations in LSM1 that inhibit BMV RNA2 translation do not affect BMV RNA3 recruitment. lsm1Δ strain was transformed with a plasmid transcribing BMV RNA3, a plasmid expressing protein 1a plus an empty plasmid or plasmids expressing LSM1 and the different lsm1 mutant alleles. Similar protein 1a levels in the presence of LSM1 and mutant alleles were achieved by using different strength promoters (GAL1 or ADH1). RNA3 accumulation was analyzed by Northern blot and detection of SCR1 was used as the loading control. The accumulation of 1a protein and Lsm1 was analyzed by Western blotting. Histogram shows average and standard errors of the mean of the relative accumulation of RNA3 from three or more independent colonies. The average RNA3 accumulation in lsm1Δ yeast expressing WT LSM1 gene in the presence of 1a protein was set to 100 ([*] P < 0.05, relative to strain expressing LSM1).
