DNA (3 nM) encoding the aptazyme and GFP was incubated in the reconstituted translation system in the presence of 0 mM (black diamonds), 0.03 mM (white squares), or 1 mM (gray circles) TPP. At the indicated times, the total RNA levels including degraded RNA (A,E), full-length RNA (B,F), cleaved RNA (C,G), and protein (D,H) were measured as described in Materials and Methods. We also performed experiments with 0.1 mM and 0.3 mM TPP, but the data used for parameter estimation in Supplemental Figure S1 are not shown here.
