Mirtron biogenesis pathway. (A) Intronic miRNAs are generated with a Drosha-dependent cleavage, while mirtrons are generated by splicing. Because splicing, unlike Drosha cleavage, does not produce defined 3′ overhangs, mirtron export may be mediated by non-exportin-5 pathways. (B) peGFP-Mirt was constructed by inserting cloning sites into a putative exon splice junction in eGFP for the introduction of mirtrons.
