Effects of the start codon and mutation A1413C on the extent of fMet-tRNA binding. Complexes were formed with f-[35S]-Met-tRNA under LS (A) and CP (B) conditions and filtered through a bilayer of nitrocellulose and nylon membranes to quantify the amount of fMet-tRNA bound. For reactions containing 30S subunits, black bars indicate wild-type subunits and gray bars indicate mutant (A1413C) subunits. Ordinate values correspond to the amount of fMet-tRNA bound per 30S and are corrected for background, which was determined from mock reactions containing f-[35S]-Met-tRNA and buffer only. Data represent the mean ± SEM from three or more independent experiments.
