EBER1 can compete with the binding of AUF1 to ARE RNA in vitro. (A,B) End-labeled TNFα-ARE RNA (A) or EBER1 (B) was incubated at 2 nM with increasing amounts (from 50 to 1000 nM) of recombinant p40AUF1. RNP complexes were resolved on a native polyacrylamide gel by electrophoresis for 2 h in 0.5× TBE buffer. (C,D) ARE RNA (2 nM) was incubated with 200 nM p40AUF1 (indicated by +), together with increasing amounts (from 5 to 50 nM) of EBER1 (C) or VAI RNA (D) as a competitor. All bandshift assays were conducted in the presence of 0.02 mg/mL tRNA as a nonspecific competitor.
