Maf1-mediated repression of RNA polymerase III transcription inhibits tRNA degradation via RTD pathway

Multicopy suppressors of trm4Δtrm8Δ defect encoding known tRNA-interacting proteins. trm4Δtrm8Δ cells were transformed with multicopy plasmid pFL44L carrying VAS1 or TEF1. tRNA^Val(AAC) gene as a positive control and empty plasmid as a negative control. Transformants were grown overnight at 30°C in SD-ura liquid media, diluted to OD₆₀₀ = 1.0 and serially 10-fold diluted, spotted on YPD plates, and incubated at 30°C or 37°C for 2 d.
