Adapter ligation to 5′-ends of RNA templates with different oligonucleotides. (A) SalI adapter ligation to 5′-ends of 5′-monophosphorylated RNAs. (B) BC1 RNA ligation with fusion SalI adapters; the SalI restriction site was exchanged for the Roche MID tags (MID1–12), the resulting linkers being Ad1–12. (C) Different RNA templates ligated to Ad1 adapter derivates. Either 3′-nucleotides or sugars were altered (Table 2). (A–C) Lanes 1 and 2 represent control reactions.
