Effect of an all-DNA stem on γ-toxin activity. The 39-mer oligonucleotide substrates are shown. The R39 substrate is all-RNA. D16•R7•D16 is a chimera in which the 16-bp stem is all-DNA and the anticodon loop is all-RNA. In D17•R2•D20 only the wobble U and the 5′ flanking U are ribonucleosides. Nuclease reaction mixtures contained 200 fmol 5′ 32P-labeled 30-mer substrates and γ-toxin as specified. The extents of 39-mer cleavage are plotted as a function of input γ-toxin.
