Identification of PUF-11 binding sites. (A) Yeast three-hybrid selection used to identify PUF-11 binding sites. The randomized region of the RNA library is illustrated using N to indicate a randomized base. Interaction between PUF-11 and RNA X will trigger expression of reporter genes HIS3 and LacZ. (B) Summary of three-hybrid selection experiment. A total of 666 positive clones were obtained. Of these, 283 clones were recovered from Escherichia coli and sequenced. Sixty-six unique sequences were present among the 283 clones, and these unique sequences yielded three classes of RNAs.
