Inhibitor potency improves with the addition of double-stranded flanking regions. Inhibitors targeting miR-21 in HeLa cells were designed with flanking regions containing polypyrimidine (A) and arbitrary (B) sequences. Inhibitors containing 8 and 16 nt single-stranded flanking regions (8Y + RC + 8Y, 8A + RC + 8A, 16Y + RC + 16Y, 16A + RC + 16A) were compared to 8 bp hairpin structures (8Yhp + RC + 8Yhp, 8Ahp + RC + 8Ahp) and 16 bp double-stranded structures (16Yds + RC + 16Yds, 16Ads + RC + 16Ads).
